Synthesis of Bufadienolide Natural Products and Analogs via Singlet Oxygen Oxidation and Late-Stage Modification
Abstract & Summary
A unified synthetic route to bufadienolide natural products and analogs was established using singlet oxygen [4+2] cycloaddition to construct the α-pyrone ring. Divergent late-stage functionalization enabled scalable access to cinobufotalin, bufotalin, and targeted structural variants for structure-activity exploration.
